The Na/K-ATPase is the transport system for Na/K-ions across the plasma membrane of most animal cells. In the plasma membrane of erythrocytes, the Na/K-ATPase is the terminal step in the 2Na(+)/1K(+)-exchange. The Na/K-ATPase is a heterodimer with a catalytic alpha-subunit and a glycosylated beta-subunit. Insulin increases the activity of Na/K-ATPase and increases its affinity for Na. Furthermore, the activated receptor forms a complex with the beta-subunit, which is dissociated from the alpha-subunit during insulin binding. The effect of insulin on the Na/K-ATPase is mimicked by H89, a protein kinase-A-inhibitor, suggesting that insulin exerts its action via activation of protein kinase-A. H89 also blocks the action of lysophosphatidic acid. The finding that a tyrosine-phosphorylation system is involved in the action of insulin on the Na/K-ATPase is further supported by the effect of tyrosine-phosphatase inhibitors. We therefore conclude that insulin affects the Na/K-ATPase via tyrosine phosphorylation. The Na/K-ATPase is also affected by a Na(+)-sucrose cotransporter, the giardin-m14 complex. The finding that giardin-m14 is inhibited by insulin suggests that the transporter interacts with the same pathway as the Na/K-ATPase. We have recently shown that giardin-m14 is activated by lipids via a GTP-binding protein, Rab-GDI. The finding that giardin-m14 can be activated via the G-protein-coupled receptor (GPCR) and via a G-protein-independent pathway suggests a role of giardin-m14 in intracellular signalling.package com.freeletics.client.response.leaderboard;
